Standardized variable is DPIP which is an electron acceptor that intercepts the hydrogen ions and electrons released from succinate, changing the DPIP from an oxidized state to reduced state. DPIP is blue in its oxidized state but changes from blue to colorless as it is reduced. DPIP is a substitute for FAD which accepts electrons from succinate to form fumarate during krebs cycle. The present experiment demonstrates the effect of succinate on state IV respiration. Magnesium sulfate is used to regulate activity of succinate dehydrogenase which converts succinate to fumarate.
