You want to produce plasmid A in bacteria. You ask your friend to give you plasmid A. He gives you the plasmid, however, your friend has digested plasmid A with a single restriction endonuclease and then has treated it with calf intestinal phosphatase (CIP). As you want to produce more of this plasmid by growing it in bacteria, before you transfect the plasmid into bacteria you will need to treat it with two enzymes. What are these enzymes, and why are you using them? Please explain your logic briefly.

For growing the plasmid in bacteria plasmid has to be traet by polynucleotide kinase and then with Ligase enzyme. Beacuse CIP catalyze the hydrolysis of 5'-phaspate group from nucleotides.
