Question
You are studying an enzyme and an inhibitor of that enzyme kinetically. A Lineweaver Burk plot of the enzyme with increasing amounts of substrate in the absence and presence of the inhibitor is shown below (Without inhibitor: x-intercept = -0.4, y-intercept = 0.2; With inhibitor: x-intercept = -0.4, y-intercept = 0.35). Calculate Km for the enzyme.
For the above question, calculate Vmax for the enzyme. Provide the numerical value and do not worry about units.
For the above question, what type of inhibition does the inhibitor work by? Provide your answer as competitive, uncompetitive, or noncompetitive.
Explanation / Answer
From the Lineweaver Burk plot
Y-intercept= 1/Vmax
So, Vmax= 1/Y-intercept
Vmax for enzyme without inhibition= 1/ 0.2 = 5
Vmax for enzyme with inhibition= 1/0.35 = 2.857
The inhibitor for the enzyme changes the Vmax on the Y-intercept, so it is non-competitive inhibitor
