Given above is the table with Rf values for some of the amino acids
As can be seen as the hydrocarbon side chain of the amino acid increases, it becomes less polar and thus moves faster on the plate, giving high Rf values.
As the polarity of the side chain increases, the amino acid interacts more strongly with the stationary phase on the plate and thus moves slower, giving low Rf values.
The amino acids with large side chains tryptophan, tyrosine and leucine. Tryptophan has the highest Rf value of all. It is very close to the Unknown spot 1 Rf value and most likely the identity of the unknown spot 1 is tryptophan.
Unknown spot 2 has the Rf more identical to aspartic. Therefore the side chain of aspartic and the unknown spot 2 might be similar.
Overall as the size of the side chain of the amino acid increases, the Rf value was also found to incease due to decreasing polarity of the amino acid.
