A non competitive inhibitor will be no match against the substrate. that means it will be atached in the enzyme in a differente site instead the substrate that will be in the active site. So the affinity will be no affected in the non competitive inhibitor but it will reduce the velocity in which the reaction happens.

Tocalculate the Km and Vmax we will need ti plot the 1/v against 1/S values. With this plot we can calculate this values with this equations.

V=Vmax([S]/Km+[S])

And

1/V=Km/Vmax(1/[S])+!/Vmax.

So with the values and the equation we calculate Km and Vmax.
