Question
chemicals used acetylferrocene term-butyl methyl ether and hexane
Please answer a b c d and e .
This is a micro scale column chromatography lab.
Briefly describe the hazards associated with the compounds used in this experiment. Why is it important to prevent air bubbles from forming in the column bed? Briefly explain why the chromatography column should be kept absolutely vertical while it is being packed and used. How will you know when you should begin to collect the acetylferrocene sample as it elutes from the column? What is the purpose of adding alumina on top of the sample?Explanation / Answer
Answer to>>a)Care should be taken in this experiment because ferrocene and acetylferrocene used as a materials are toxic .The clourless tert butyl methyl ether has a distinctive anesthetic - like odor and vapours are heavier than water and it is narcotic. It has a effective effect on increasing serum albumin and total proteins in rat when they consumed it highly.
Answer to>>b) Air bubles present in the column bed cause the bed irregular i.e.,column contains gaps where no solid phase is present and interfere with the uniform movement of the mixture of the compounds. This will decrease in the resolutin of the separation that is lead to a decrease efficiency of the separation.
Answer to>>d)As the solvent goes down the column in chromatography experiment then you will prprobably seen a yellow colour band followed by an orange colour band .A orange colour band at the bottom of the column will come, which confirms elution of acetylferrocene from the column.
Answer to >>e)When alumina adds on the top of the sample then it will prevent disruption of the sample layer with addition of eluent.
