The drug has the absorbance in the UV region which can be used to estimate the concentration of the drug. Using Beer Lambert equation, we can estimate the concentration of the substance (drug).

3. We can use two solvents, one of the them being water and other phase (hexane or any other solvent, based on the polarity required) is organic in nature in a separating funnel. So when we take the drug in aqueous phase and equilbrate with the organic phase, there is a distriburiton of the drug in two phase which we define the partition coefficent (The concentration of the drug in the organic phase to the concentration in the aqueous phase). We can estimate the drug in the aqueous and organic phase by measuring the concentation of the drug by using UV spectrophotmeter. Hence we can estimate the equilibrium concentration for a given pair of solvent for the drug.
