The difference between in vitro and in vivo release rate of drug is due to non degradable biomaterial used. During in vitro drug delivery on thin slab this non degradable biomaterial is unaffected by other chemicals or enzyme due to controlled nature. But in case of in vivo drug delivery, this non degradable biomaterial is affected by many chemicals and enzymes of the body and undergoes some changes that prevent release of drug in due course of time. That's why the graph of in vivo release of drugs shows zero release after 12 days.
