Question
Dr. Blake has given you a flask containing HepG2 cells and DMEM media. He has asked for you to perform a series of serial dilutions with the cells to plate for a series of experiments. After counting the cells you determined the stock concentration to be 5.0 x 10E7 cells/mL at 9mL and can now dilute the stock into two solutions. Additionally, he has asked to make sure you have enough in each solution to use to plate so be sure to not use all of it . Perform the serial dilution with a minimum of two steps each.
A:
- Concentration = 2.5 x 10E5 cells/mL
-Volume = 50 mL
B:
-Concentration = 1.0 x 10E5 cells/mL
- Volume = 50mL
Please explain step by step how you got your answer!!!!!
Explanation / Answer
HepG2 cells are perpetual cell lines derived from liver tissue. They are hepatocellular carcinoma cells, and can grow on DMEM (Dulbecco’s modified Eagle’s Medium) media. The stock solution, which will be again diluted to lower concentrations, contain 5.0 x 10e7 cells/mL at 9mL.
a. Suspend this solution in 50 mL of DMEM media, 5.0 x 10e7 cells/mL x 50 ML = 2.5 x 10e9 cells.
b. Resuspend the solution obtained in another 50 mL of DMEM media, 2.5 x 10e9 cells/mL x 50 ML = 1.25 x 10e11 cells total.
