Question

The Poisson distribution analysis can be used to calculate the titer of a given suspension of virus. I ran an experiment dealing with egg infectious dose using a newly isolated paramyxovirus in suspension. I would like to know the titer of the virus suspension in egg infectious dose units. 100 eggs were inoculated with ten microliter of a 1: million dilution of the virus suspension. After five days of incubation I discovered that ten eggs were not infected.

Calculate the titer of my virus in the undiluted suspension. What are the units ?

Explanation / Answer

titre(pfu/ml) = number of plaques/(dilution*volume of diluted virus added to the egg)

titre= 90/(0.000001*0.01)

titre=9*109 pfu/ml
