Question

The Poisson distribution analysis can be used to calculate the titer of a given suspension of virus. After several attempts to grow the agent you isolated from week # 1 it was concluded that the pathogen was indeed an enveloped, negative strand RNA virus. You succeeded in growing this virus in embryonated quail eggs from which you harvested 10 mL You would like to know the titer of the virus suspension in egg infectious dose units (EID).

To determine the virus titer, 100 eggs were inoculated with ten microliter of a 1: million dilution of the virus suspension. After five days of incubation you discovered that discovered that ten eggs were not infected at all.

Define (explain) each bold word or phrase by telling me what it means to you.

Calculate the titer of your virus (EID/mL)   in the undiluted suspension.

Explanation / Answer

Hi,
Poison distribution is a statistical measure of variables and their distibution. Events with discete probability and which can occur independently are considered for poison distribution. For example, the no. of people arriving to train station between 10 - 11 AM. This does not depend on the people who came before or after; the distributin also is independent meaning the people who come to station independently and do no influence each other.
Pathogen is a microorganism which can cause illness. A pathogen can be a bacterial, virus or prtozoa. Pathgens attack the host, cause several symptoms and illness. If untreated infections can have high rate of mortality.
Enveloped virus: A virus particle with a envelop or cover over its body. This cover is derived from the host during budding off process.
negative-strand RNA virus: Negative strand means non template strand. It cannot code for mRNA. After infection, the negative strand is converted into positive in order to produce mRNA.
Emryonated eggs are the growing eggs. Thes are used to culture virus for research. These eggs are in advanced stage of development.
Titre is the number of viral particles in the solution.
egg infectious dose units (EID) is the amount of virus required to infect 50% of the eggs.

The EID/ml = [(total number of animals died/number of animals inoculated per dilution) + 0.5] × log dilution factor.
= [(90/100) +05] x 10^6
= 1.4 x 10^6 /ml
