A company approaches your supervisor with a new drug that has great anti-tumour properties and also inhibits the growth of yeast. The company wants to understand the mechanism of action of the drug. You perform a number of experiments in model organisms. How would you interpret the following results

a. A screen of the non-essential yeast deletion collection with the drug leads to a fitness profile similar to those reported for drugs that are known to induce production of reactive oxygen species.

b-A haploinsufficiency screen identified an rbp5/RPB5 which has a heterozygous deletion for RNA polymerase B as sensitive to the drug. What does this tell you about the potential drug target?

c-What model organism would you use to look at toxicity of the drug?

Explanation / Answer

a) ROS producing drugs reduce the fitness of cells by activating apoptosis. Since the drug effects similar to ROS activating drug, it means the drug is cytotoxic.

b) RNA polB becomes sensitive to the drug, it may affect general transcription of the cell by binding to RNA polB or affecting its assembly.

c) C.elegans is suitable for studying apoptosis and transcription impact in vivo by the drug.
