Examine the leaf first with the low-power objective to gain an overall impression of its morphology. Note 3. Find the centrally located midvein within the midrib, the midvein-supporting tissue. You might think of the size and orientation of the veins within the leaf. The veins contain the xylem and phloem. the midvein as the major pipeline of the leaf, carrying water and minerals to the leaf and materials pro- duced during photosynthesis to sites where they will be used during respiration. Use the high-dry objective to examine a portion of the blade to one side of the midvein. Starting at the top surface of the leaf, find the cuticle, a waxy, water-impervious substance covering the upper epider- mis. The epidermis is a single layer of tightly appressed cells that are tightly bound together, without any intercellular spaces. 4. 189

The present activity aims to visualize and understand the anatomical cross-section of the dicot leaf. The activity aims at collecting fresh dicot leaves from a plant and prepare a cross-section of the leaf on a microscopic slide. This slide is then placed under a bright-field microscope and focussed to visualize the anatomical structures. Various features of the leaf are observed.

Starting from upper epidermis, the mesophyll layer of cells is visualized. Deeper in the tissue, the bundles of xylen and phloem tissues can be clearly visualized in the vein section of the leaf. This means that the phenotypic apperance of the vein in the leaf represents the anatomical location for vascular bundle as well. The typical features of the cuticles, epidermis, mesophyll and vascular bundle can thus be discussed.
