You are pretty sure that these two primers will w in Bearded Dragons, so you decide to go ahead with the PCR. In frogs, the Forward Xf primer binds gene Xf between nucleotides 32 and 53. The Reverse Xf primer binds between nucleotides 642 and 663. When you analyze the PCR reaction you set up using Bearded Dragon DNA, your PCR product is 700 bp. Based on this information, answer the following: you ork a. What size PCR product would these two primers create if they were used to amplify the frog gene Xf?? Forward primer NT: 5' 3" Reverse primer NT: PCR Product Size bp b. Based on this predicted frog PCR product and the Bearded Dragon PCR product you created, do you think that these two primers amplified gene X in the Bearded Dragon? Why or why not?

F-Xf binds between nucleotides-32 & 53

R-Xf binds between nucleotides- 642 &663

So the PCR amplified product in frogs with these primers have a size of- (32-663) = 631 bp (Answer a-631bp)

Answer-(b) But when the same primer used to amplify Bearded Dragon DNA the amplicon size was 700bp i.e.69 bp extra was found in comparison to Frogs. So, I don’t think that these primers amplified the gene X in Bearded Dragon because if the gene is similar than amplicon size should be similar to frogs amplicon size i.e 631 bp.
