Question

A cell is deficient in pyruvate dehydrogenase phosphate phosphatase. How would such a deficiency affect cellular metabolism? Lipoic acid and FAD serve as prosthetic groups in the enzyme isocitrate dehydrogenase. Describe their possible roles in the reaction catalyzed by the enzyme. Explain why the metabolism of a C_15 fatty acid can lead to the net synthesis of glucose, but the metabolism of a C_16 fatty acid cannot. What, if anything, is wrong with the following statement: "The oxygen consumed during the oxidation of glucose in animal cells is returned as CO_2 to the atmosphere." How might you support your answer experimentally?

Explanation / Answer

Pyruvate dehydrogenase complex made up of three components E1, E2 and E3 which convert pyruvate to acetyl CoA, an important step of energy metabolism which enters in kreb cycle. E1 refers to pyruvate dehydrogenase and present in cell in two forms active and inactive. Pyruvate dehydrogenase phosphatase dephosphorylates the inactive form of E1 to active forms.
