restriction enzyem cuts the BS allele in normal individual. it indicates that if the person is normal than the mutation in sickle cell anemina absent. as we know that there are two allele for a particluar gene hence in case of stolla and tonia one allele which is normal digested with MstII gives two bands and one mutant allele is not digested with the same enzyme and it will remain intact. so there are three bands observed in case of carrier (stolla and tonia). Tim and Joe are homozygous normal individuals becasue, they do have present restriction site on both alleles hence gives two bands only.
if any of the individual in this family is affected, than only one band will be seen in the gel (because both the allele remain intact, non cutting site is present on any of the allele ).
