Question
6. Consider a colony of coli cells in which the gene for the CRP protein is deleted. Describe the effect of that this deletion will have on the arabinose operon in terms of Ara A protein levels when the cells are grown in media containing arabinose but no glucose. Explain your reasoning. (3 points) 7. Draw what the results for a Northern blot, probing for ara a mRNA, would look like for coli cells grown under the following conditions: 1) arabinose absent and glucose present, 2) arabinose present and glucose absent, 3) arabinose present and glucose present. Explain your reasoning. (4 points) 8. Draw what the results for an EMSA would look like for the following samples: 1) CRP CAMP lac operon DNA sequence, 2) CRP lac operon DNA, 3) LacR + allolactose lac operon DNA, 4) LacR+lac operon DNA, 5) AraCarabinose ara operon DNA sequence, 6) AraC+ ara operon DNA sequence. For you drawing, assume that the lac operon DNA sequence and ara operon DNA sequence are approximately the same size. CRP dimer weighs 84 kDa, LacR tetramer weighs 154 kDa, and the AraC dimer weighs 72 kDa. (3 points)Explanation / Answer
Ans 1) The CRP gene is one that belongs to the pentaxin family and it is able to recognize many foreign pathogens along with damaged cell in the host. It works in an attempt to eliminate the foreign and damaged cells from the humoral and effector system in the blood. The gene is responsible for many host defense function that leads to many function like agglutination, capsular swelling, phagocytosis etc.
Arabinose operon is a very important regulation process and it is required for breakdown of the 5 carbon sugar L-Arabinose in E.Coli. Ara A is one of the structural genes that encodes isomerase. In this case when the cell are grown in a media containing arabinose and no glucose they need to gain the energy for growth from arabinose by breaking it down. However, if the gene for CRP is deleted, there will be no CRP and hence it will interfere with the function of the structural genes. It was found that if there is a mutation in the CRP gene, it impacts all the structural genes but not AraA. However it complete deletion is also going to slow down the process of Arabinose breakdown for gaining energy.
