In any signalling process a signal molecules comes from a site where it was produced and then make interaction with targate cell.

Most of the cells have membrane bound receptors on which these signalling molecules binds and do funther multistep signal transduction process. After complete processing cell produce a responce against the processed signal.

So signal comes first and binds to the receptor and signal transduction is proceeded in cytoplasm and then cell show response against signal.
