Question

Laboratory Report 18 18 Motility Determination A. Results 1. Which bacterial species exhihited true motility on the slides? 2. Draw the appearance of the inoculated semisolid or SIM tabes. Did the semisolid medium inoculations concur with the slide results Micrococcus lutews Proteus vulgaris 3. Descrihe the differences hetween the two stabs in the sot agar plate. Based on these resulis,w hich species appears to be motile? B. Short-Answer Questions 1. Describe the structure of a flagellum. How do flagella generate cell moxtility? If you compared two motile bacterial species and determined one was considerably more motile than the other, which arrangement of flagella would you expect to be associated with the highly motile species? How would you confirm this? 2. 133

Explanation / Answer

Answer 1) proteus vulgaris

2) yes, P. vulgaris appeared as motile in the slab tubes.

Answer b) flagella has 3 parts:

Flagella are helical shaped structure which is composed of subunits of a protein called flagellin.

The wider region at the base of the flagellum is called hook.

Hook connects filament to the motor portion of the flagellum called basal body.

The movement of eukaryotic flagella depends on adenosine triphosphate (ATP) for energy, while in prokaryotes flagella derives its energy from the proton-motive force, or ion gradient, across the cell membrane.

2)Monotrichous flagellar arrangement has less likelihood of hangling, so it will cover greatest distance per unit of time.

for conformation of this, inoculate both culturs into stab tubes and examine which moves from the stab line at the fastest rate.
