You are using 1 microlitre of the enzyme means 20 units of it. This digests your yeast genomic DNA in 2 hours. 1 unit of the enzyme means that you can digest 1 microgram of DNA with it. So ideally you can digest 20 micrograms of DNA in 1 hour. Since it takes 2 hours for complete digestion, the DNA content probably is 40 micrograms.
The next part requires the amount of cell culture you have taken for digestion. Supposedly, you had taken 1 ml of your culture for digestion. Then in that 1 ml you had 40 micrograms of DNA. Since 1 ml of culture contains 20 million cells, these cells have 40 micrograms DNA. Applying unitary method yields the DNA content per cell as 2 nanograms.
