The total number of nitrogen bases in DNA (A, T, G and C) are four. So, at a time one can accomidate at one posistion, so the proportion of one base at one position is (Either A or G or T or C) = 1/4.

As the restriction site has 4 nucleotides length, the probability of occurance of the restriction site = 1/4 x 1/4 x 1/4 x 1/4 = 1/256.
