Hydration medium plays a very important role in functioning of liposomes. Hydration medium should of same similar osmolality to the buffer/medium of that used in liposomes to avoid osmotic effects. If the medium contains high concentration of salts, liposomes start to precipitate. This is because hydrating medium influences zeta potential of the liposomes. The liposomes present in the medium such as deionized water possess high zeta potential as compared to those present in solutions such as phosphate buffer. Zeta potential influences the packing of the lipids in bilayer. A greater colloidal stability is noted in deionized water. Change in the hydration medium can alter diameter of the liposomes, affect encapsulation and rate of drug release. During active drug loading in liposomes osmolality within liposomes has to be different from that of outside to facilitate drug diffusion. In case of liposomes prepared from from phospholipids, PBs buffer of ph 7.4 and ionic strength upto 100 mM of sodium or potassium phosphate buffer saline is recommended.

Hence the choice of the solvent for self-assembly of lipids to form liposomes needs to be taken critically as it affects to self-assembly of lipids which is based on the hydrophobic effect between hydrophobic tails. Liposome hydration or suspension media such as buffer or saline solution or isotonic sucrose solution should be chosen carefully as these affect the properties of lipid that influence the formation of liposome. The ions concentration may be effect on stability of nano liposomes with electrostatic repulsion. Non-ionic solution can be used for hydration.

The choice of solvent during hydration also depends upon the drug or compound that needs to be encapsulated in to liposomes. Suitable hydration media include distilled water, buffer solutions, saline, and nonelectrolytes such as sugar solutions are the accepted solutions. These include are 0.9% saline, 5% dextrose, and 10% sucrose. During hydration some lipids form complexes unique to their structure. Highly charged lipids have been observed to form a viscous gel when hydrated with low ionic strength solutions.

Precipitation in the confinement of liposomes excludes heterogeneous nucleators and facilitates formation of the thermodynamically most stable crystalline phase. According to the question, the cystal of NaCl should be formed within the liposome.
