Question
Liposomes containing specific (one kind only) containing v-SNRES and t-SNARES were reconstituted. To measure fusion, v-SNARES liposomes contained a fluorescent lipid Fluorescence was quenched until fusion of liposomes happened. Shown below is the data when liposomes containing a particular v-SNARE were fused with a particular t-SNARE. Answer the following questions; 2. -SNARE 1 v-SNARE 2 v-SNARE 3 tSNARE = plasma membrane tSNARE = Golgi tSNARE = vacuolar Time after mixing Where would you expect v-SNARE 1, 2 and 3 to be localized in the cell?Explanation / Answer
Vescilce and membrane can only fuse when there is a complementary combination of v-SHARE on the vesicle and t-SNARE on the recipient membrane.
In case of upper panel fluorescence, the intensity is increased when v-SNARE 1 and 3 fused with the t-SNARE of Plasma membrane. So v-SNARE 1 and 3 are localized to the plasma membrane.
In case of middle panel fluorescence, the intensity is increased when v-SNARE 2 fused with the t-SNARE of Glogi. So v-SNARE 2 is localized to the Golgi.
In case of lower panel fluorescence, the intensity is increased when v-SNARE 3 fused with the t-SNARE of Vacuole. So v-SNARE 3 is localized to the vacuole.
