Ans a) P 56 was found to inhibit parasite of malaria cysteine protease and it have the potential to be used as an antimalarial drug.the mechanism of action of p56 as dock protein downstream of receptor or non-receptor tyrosine kinase.
So p56 act as a competitive inhibitor because as show in figure Km is increased and the Vmax remain the same.
Ans b)
for Inhibitor P56-
the KM = -(1 / -0.5µM-1) = 2 mg/ml
and Vmax = 1 / 2.35 mM-1*s = 0.425 µmol/µl
for No inhibitor-
the KM = -( 1 / -1.2µM – 1 ) = 0.83 mg/ml
and Vmax = 1 / 2.25 mM-1*s = 0.44 µmol/µl
Ans c) Calculation for the value of Ki in the 0.22 mM inhibitor presence .
becoz 2 mg/mL / 0.83 mg/ml= 2.40 = alpha
and Ki = 0.22 mM / 2.40 -1 = 0.1571 mM
