Question

please answer the whole question

• List the subunits of which prokaryotic RNAP is composed and briefly describe the function of each subunit.

o What is the difference in subunit composition of the RNAP core enzyme and holoenzyme?

• Define the sense (coding) and the antisense (noncoding) strands of DNA regarding the process of transcription.

o Which strand acts as the template for the synthesis of RNA?

o What is meant by the terms “upstream” and “downstream” with regard to the process of transcription?

Explanation / Answer

core enzyme has 5 subunits which include-

a. ' subunit- largest subunit, makes the active center where RNAP interacts with DNA sequences.

b. subunit- second subunit, adjacent to ' subunit and comprise of rest of active center

c,d. subunit- two alpha subunits are present in each copy of RNAP. it contains 2 domains i.e., C terminal and N terminal. it is involved in recognizing promoter sequences and other factors.

e. subunit- smallest subunit, facilitates assembly of core enzyme.

the composition of core enzyme constitutes stated subunits, whereas holoenzyme consists of another subunit called sigma factor which helps to initiate the process.

coding strand is the strand whose sequence is similar to the transcribed RNA strand and the template is called antisense strand.non-coding strand acts like the template for RNA synthesis.

5 prime end of DNA is downstream and 3' end is upstream for DNA molecule and for an RNA it is exactly molecule.
