The following experiment was conducted such that the same amount of starch was added to test-tube A, test-tube B, test-tube C, and test-tube D were placed. Additionally, in test-tube A and D, amylase was added at the same time. Based on the above experiment, answer the following questions: 1. In which test-tube will there be the highest amount of glucose? Why would the test-tube that you selected contain the highest amount of glucose? 2. What would happen to the enzyme if sulphuric acid was added to test-tube D?

Explanation / Answer

1.Highest amount of glucose would be present in Test tube D.This is because we know that amylase works best in human body at a temperature of 370c which is normal body temperature.This condition is satisfied in tube D which is placed in water bath at 37oC.So amylase would break down starch to produce high amount of glucose.In case of tube A, the temperature of water bath maintained at 15oC would not allow for efficient functioning of the amylase.

2.If sulfuric acid is added to test tube D, it would denature the amylase bringing about a change in its configuration and shape such that it would no longer be able to carry out its function.Thus, no starch will be hydrolysed to glucose in test tube D.
