If we have 4 exon means Exon 1, Exon 2, Exon 3 and Exon 4. In this Exon 1 and Exon 3 join together and do their function for specific compound. So we have to see onlt this mRNA codon. So we have to take cDNA from this mRNA for the amplification not the whole genome DNA.

2. Ori site is missing. Ori site is important for replication orgin. WIth out this no replication happen in the vector. So, only one copy is transfered to one cell and other cell is not have any vector
