Question

Southern Blot Exercise 1 20f 10 ) An individual with the genotype H1H3 was analyzed by Southern blot using Probe A. Which of the following is the correct description of the expected result? 7 .0 Two bands, corresponding to 4 kb and 7 kb restriction fragments A band corresponding to a 8 kb restriction fragment. A band corresponding to a 4 kb restriction fragment. Two bands, corresponding to 3 kb and 5 kb restriction O 8.0 Probe Probe fragments A band corresponding to a 7 kb restriction fragment. Two bands, corresponding to 3 kb and 8 kb restriction The figure above shows the restriction map of four alleles of the H gene (H1, H2, H3 and H4). The arrows indicate the position of sites that can be cut by the restriction endonuclease, EcoRI. The numbers indicate the distance between sites in kilobase pairs (kb). The shaded regions labeled Probe A and Probe B show the location where single stranded DNA probes would hybridize (base pair with) gene H DNA. Probe A is complementary to the shaded region labeled Probe A and Probe B is complementary to the shaded region labeled Probe B. fragments A band corresponding to a 3 kb restriction fragment. Two bands, corresponding to 3 kb and 7 kb restriction fragments Two bands, corresponding to 2 kb and 5 kb restriction fragments Each of the questions below ask you to use this figure to determine the results of Southern blot anlayses of the H gene in individuals with various H genotypes (for example, H1H2 is an individual heterozygous for H who has one H1 allele and one H2 allele). In each Southern blot analysis, DNA is isolated from the individual digested with EcoRI such that all EcoRI sites are cut, the DNA is run on a gel, denatured, transferred to a membrane, and the membrane is probed with either Probe A. Probe B, or both. Refer to the information in the quesitons and in this Two bands, corresponding to 2 kb and 8 kb restriction 0 fragments A band corresponding to a 2 kb restriction fragment.

Explanation / Answer

The correct answer is a band corresponding to a 4 kb restriction fragment because from H1 there will be four bands those are 2kb, 3kb, 3kb, 4kb and from H2 the bands will be 5kb, 3kb and 4 kb. When the probe A is used then according to the figure it will only bind to the 4kb region of H1 and H3. That is why only one band will be observed.
