1) What is the total dilution factor of a serum sample if you:

a) add 5ml of serum to 30ml of saline?

b) add 1ml of serum to 99ml of saline?

2) You have 4ml of packed SRBCs. Using all of the SRBC, how much saline do you have to add to make a 25% suspension of the SRBCs? How much total volume of the 25% suspension will you have?

3) You made a 1/2 dilution of a serum sample. Then you added 2ml of the diluted serum to the 4ml of water. Then 1ml of that was diluted to a total of 10ml. What is the total dilution factor of the serum?

4) You took 6ml of a glucose solution and diluted it to a total volume of 20ml. Then a 1/15 dilution was made using the diluted glucose solution. What is the the total dilution factor?

5) You have a 1000mg/dl solution of sucrose. You took 1ml of that and diluted it to a volume of 4ml. Then you took 4ml of that and added it to a 36ml of water. What is the final concentration of your glucose solution?

Explanation / Answer

Dilution factor = Final volume/initial volume
1.
a.
5 mL of serum in 30 mL of saline
= 35/5
= 7

b.
1 mL of serum in 99 mL of saline
= 100/1
= 100

2.
25% solution = 25 mL in 100 mL
Initial volume = 4 mL
Final volume = (4 X 100)/25 = 16 mL
Volume of saline to be added = 16 - 4 = 12 mL

3.
Starting concentration = A
1/2 dilution = A/2
2 mL of A/2 is mixed with 4 mL of water.
Dilution factor = 6/2 = 3
Total dilution factor = 2 X 3 = 6

1 mL of the above dilution is diluted to 10 mL
Dilution factor = 10/1 = 10
Total dilution factor = 6 X 10 = 60

4.
6 mL of glucose diluted to 20 mL
Dilution factor = 20/6 = 3.33
From this, 1/15 dilution was made.
So, 1 mL of this dilution is made upto 15 mL
Dilution factor = 15
Toatl dilution factor = 3.33 X 15 = 50
