Explanation / Answer

1. In size exclusion chromatography molecules are separated by their size and in some cases molecular weight .It is used to separate biological molecules like protein.

2. In size exclusion chromatography mixture of molecules dissolved in liquid is applied to column which contain beads or stationary phase. This act as sieve or trap and function to filter small molecules ,large molecules pass or excluded from the beads thus large molecules elute first while small molecules get trapped.

Stationary phase is a semipermeable porous beads they are cross linked polymer of smaller pore size are used for rapid desalting of protein .Stationary phase must be inert ,inexpensive and should not react with eluent.
