Question
Explain in full detail please!
A 180 nucleotide segment of the nontemplate (mRNA-like) strand of a eukar-yotic protein-encoding gene is shown below. The inferred amino acid sequence of the three forward reading frames are shown below the sequence. * indicates an in frame termination codon. This sequence spans two exons and the intron between them. Based on what you know about spliceosomal introns, predict (1) the location of the 5*(donor) splice site, (2) the location of the 3' (acceptor) splice site, and (3) the probable location of the branch site. Write the amino acid sequence of the protein that corresponds to the two exons.Explanation / Answer
Answer:
In the 180 nucleotide segment given in the figure, the location of the 5' splicing site would be at the 1st line (predicted protein sequence in first line) is at the nucleotide potion 70 where you get TAA i.e UAA which is a termination codon. The sequence at this is the 5' doner site and
at the 3' acceptor site of the first line of amino acid is TGA i.e UGA , the point which is also termination codon.
The splicing of the intron from nucleotide 70 to 120 will give the protein which contains the aminoacid as
L V I C G T N R L K P L C R T Y A F M P Q R V S L L L G W E S T N L R K K.
At the end of this again stop codon is there at 3' end. so the above will be the sequence of protein which contain two exons.
