Question
Shown below are the amino acid sequences of the wild-type and three mutant forms of a short protein.
A fourth mutation in the gene is isolated. Analysis of the new mutation shows that the protein encoded has the wild-type sequence, but cells produce 50% more protein than in wildtype cells. Where would you predict the mutation is located in this strain?
Wild type: met-trp-tyr-arg-gly-ser-pro-thr
Mutant 1: met-trp-tyr
Mutant 2: met-trp-tyr-arg-ala-leu-ala-gln
Mutant 3: met-trp-his-arg-gly-ser-pro-thr
Explanation / Answer
Mutant 1- deletion or non-sense mutation whree an termination codon appears between the gene resulting in truncated protein,, as the no of amino chain is short
Mutant 2 - inversion as the amino acids is same but different sequence hence the gene is inverted hence coding changes..
Mutant 3- substituton mutation whre the base is changed resulting in the formation of the different codon hence different amino acid coding.
Mutant 4- the mutation must be in the Supressor gene which prevents the suppression of the gene thus preventing the expression of the genetic due to which the gene gets highly expressed without inhibitions...
