Question
Hi, I am trying to answer my lab questions on bacterial transformation. I select the answers for each of the questions already. Can someone help me check if the answers are correct or not. Quick question: for this type of lab how do you determine which plate had the most bacterial growth? Really appreciate your help!
Plate 1: No pGLO w/ broth
Plate 2: No pGLO broth+ampicillin
Plate 3: pGLO broth+ampicillin
Plate 4: pGLO broth+ampicillin+arabinose
1. On the pGLO plasmid, what is the ori sequence for? A) It is the origin of replication so the bacterial cell can copy the plasmid. OB) It allows us to select for bacterial cells that picked up the plasmid. OC) It codes for the green fluorescent protein OD) It allows us to control whether the GFP gene is expressed or not.Explanation / Answer
10. B. The GFP was made but was inactivated because of the heat. The GFP protein is not thermal resistant gene, when heated it get inactivated upto 80%, research have been undergone on this inactivation,, due to presence of pGLO plasmid containing GFP it will produce but it is heat inactivated hence no band is observed.
