miRNA and siRNA both binds to mRNA in different ways and leads to its degradation. As a result of it, mRNA is not able to translate itself into protein.

When the cells were treated with siRNA and miRNA, leads to degradation mRNA.

Northern blot detects the presence of mRNA when radiolabelled P32 primers (specific for the gene of interest) were hybridized with the total RNA of the cell. Given miRNA and siRNA were specific against the gene of interest. Upon treatment with miRNA and siRNA, mRNA degraded and there was no band in the northern blot whereas in the case of the control sample cells were treated with either with scrambled miRNA of no miRNA or siRNA due to which one band was visible in the northern blot.

Western blot is a technique used to detect the expression of proteins inside the cells. In western blot, specific antibodies are used which binds only to the protein against which they are developed.

In the given condition when cells were treated with miRNA or siRNA (which are specific for the gene of interest), leads to degradation of mRNA and no translation of the protein of interest. As a result of it when the isolated protein was subjected to western blotting there was no band of the gene of interest. In case of control, sample cells were treated with either with scrambled miRNA of no miRNA or siRNA due to which one band was visible in the western blot
