Question
You will prepare three samples of DNA A and three samples of DNA B. The total volume of each of the samples needs to be 12ul, but it is up to you to decide what fraction of those 12 ul is your DNA sample. You can choose to do the same volume of DNA in each sample, or different volumes. Group "Brave Molecular biologists" decided to have sample A2 contain twice as much DNA as sample A1, while sample A3 will contain twice as much DNA as sample A2. The loading dye (sample buffer) is provided at 6X concentration, meaning that it needs to be diluted 6 fold (so dilution factor is 6, remember those straight dilutions!). Which of the following is the correct set up?
A. Sample A1: DNA = 2ul, loading buffer = 2ul, water = 8 ul
Sample A2: DNA = 2ul, loading buffer = 2ul, water = 4 ul
Sample A3: DNA = 2ul, loading buffer = 2ul, water = 2 ul
B. Sample A1: DNA = 2ul, loading buffer = 6ul, water = 6 ul
Sample A2: DNA = 4ul, loading buffer = 4ul, water = 4 ul
Sample A3: DNA = 8ul, loading buffer = 2ul, water = 2 ul
C. Sample A1: DNA = 2ul, loading buffer = 2ul, water = 8 ul
Sample A2: DNA = 4ul, loading buffer = 2ul, water = 6 ul
Sample A3: DNA = 8ul, loading buffer = 2ul, water = 2 ul
D. Sample A1: DNA = 1ul, loading buffer = 4ul, water = 7 ul
Sample A2: DNA = 2ul, loading buffer = 8ul, water = 2 ul
Sample A3: DNA = 4ul, loading buffer = 12ul, water = 0 ul
A. Sample A1: DNA = 2ul, loading buffer = 2ul, water = 8 ul
Sample A2: DNA = 2ul, loading buffer = 2ul, water = 4 ul
Sample A3: DNA = 2ul, loading buffer = 2ul, water = 2 ul
B. Sample A1: DNA = 2ul, loading buffer = 6ul, water = 6 ul
Sample A2: DNA = 4ul, loading buffer = 4ul, water = 4 ul
Sample A3: DNA = 8ul, loading buffer = 2ul, water = 2 ul
C. Sample A1: DNA = 2ul, loading buffer = 2ul, water = 8 ul
Sample A2: DNA = 4ul, loading buffer = 2ul, water = 6 ul
Sample A3: DNA = 8ul, loading buffer = 2ul, water = 2 ul
D. Sample A1: DNA = 1ul, loading buffer = 4ul, water = 7 ul
Sample A2: DNA = 2ul, loading buffer = 8ul, water = 2 ul
Sample A3: DNA = 4ul, loading buffer = 12ul, water = 0 ul
Explanation / Answer
I believe the correct answer to be;
C. Sample A1: DNA = 2ul, loading buffer = 2ul, water = 8 ul
Sample A2: DNA = 4ul, loading buffer = 2ul, water = 6 ul
Sample A3: DNA = 8ul, loading buffer = 2ul, water = 2 ul
due to the fact that loading buffer (6x) is needed to be 2uL in 12uL solution to make it 1X that is supposed to be used. Moreover A2 is twice than A1 and A3 is twice than A2. Rest is made up with water upto 12uL.
