6. Glutamate oxaloacetic transaminase-2 (GOT-2) is a mitochondrial enzyme that synthesizes glutamate from aspartate and alpha-ketoglutarate. GOT-2 is a homodimer, a protein made of paired identical polypeptides. The Got-2M mutation introduces a single amino acid change that alters the charge of the polypeptide produced by the normal Got-2+ allele. When enzymes from Got-2t Got-2t homozygotes, Got-2t Got-2M heterozygotes, and Got-2M Got-2M homozygotes are separated by charge using gel electrophoresis, the gel shows the following pattern of bands Got-2+ Got-2 Got-2 Got-2Got-2 Got-2 Compared to the normal GOT-2 polypeptide, is the polypeptide produced by Got-2M Got-2M mutants more basic or more acidic? a. b. A. More basic - Basic amino acids are +ve chharge so it repel at +ve side and migrate faster towards -ve side.
B. Got2+/Got2+ - no mutation it is more -ve charge so it present in top layer. Got2+/Got2m - In this one mutant and other is wild type, so, net charge is zero. So it presence between + and _ ve end.
It is more intense because it has lot of homodimer.
SOme light bands also visible they are due to separation of homodimer. But the quantity is less.
Got2/Got2m - Both gene are mutant and conver to more basic. so it present in bottom of the gel.
