Question
F) You are investigating a small peptide that you hypothesize may be an inhibitor of the cell oyle. If this compound works as you intended, it would not kill cells, but prevent them from dividing, Drugs with similar properties are useful in research and medicine. In a quick test, you combined the peptide with human fibroblast cells in the appropriate growth media. After 24 hours, the number of cells had only increased by 30%. Normally the number would be higher, without controls, what can be concluded? What controls should have been included? Describe the appropriate negative and positive controls. (4 pts)Explanation / Answer
Without controls both can be conclusive as you cannot say confidently whether this reduction in cell number is defect of any accidental variation in the growth media and or growth condition which can be ruled out only after comparing with proper controls.
Negative controls - fibroblast cells in normal medial without addition of any drug. (In this case a normal increase in the number of cells should be obtained in the given conditions after 24 hrs- expected to be higher than the test ones in which the peptide was added)
Positive control - fibroblast cells in normal media with a known mitotic inhibitor for eg. vincristine. One must expect to get no growth or highly reduced increase in the number of cells after 24hrs.
