Q4/ Answer the following questions: A- You wish to clone pices of human DNA cut with the Res. endonuclease Bam HI into the Bam HI site of pBR322. You cut both human DNA and pBR322 with Bam HI and ligate them . You transform the ligate mix into E.coli selecting for Amp. Outline how you would determine which of the transformation probably contain a plasmid with a human DNA insert. B- Genomic clones of the chloroplastic glutamine synthetase gene (gln2) of maize are cleaves into two fragments by digestion with restriction endonuclease Hind III , where as full length maize gln2 cDNA clones are not cut by Hind III. Explain these results

Explanation / Answer

Genomic clones are the DNA fragments that is comprised of the complete genome, and in other hand, cDNA clones are prepared from the mRNA using a set of reactions. As HindIII unable to cut the cDNA clone, so it is clear that the restriction site of HindIII presents in the intron segment of the gln2 gene. Therefore when the mRNA of gln2 is produced then the restriction site is spliced out, and as a consequence, it is not present in the cDNA clone.
