In the past lab or twoyou sent in DNA samples for sequencing. In the past when the data came back a number of students had received a message from the sequencer that theirdata contained “multiple signals”. What is interesting about this is that it typically occurred on the T7 read and not the pKKrev read. As a case in point one read came back with a read that had multiple signals startingat 106 bp into the T7 read. The reverse read on the same sample, using pKKrev as the primer, did not have multiple signals at all. How can this data be explained? Basically the question is why does a T7 read have multiple signals and pkkrev does not usually have multiple signals
Explanation / Answer
The possible reason for getting multiple peaks with T7 could be due to the presence of more than one primer binding sites for T7. This can be avoided by using a different primer.
