Date b Questionsnms -Watures cetays+s 1. List any specific safety or disposal considerations for this lab. 2. How do you maintain the temperature of a water bath using tap water? 3. What observation indicates that fat has been hydrolyzed? 4. What observation indicates that a protease enzyme is active? 5. What observation indicates a positive result for a Benedict's test? 6. What observation indicates that urea has been hydrolyzed? 7. What substance in the above question is responsible for the observation? catalase? 8. What observation is used to detect the activity of EnzymesExplanation / Answer
1) Enzyme work in optimum temperature, pressure ,concentration and pH.So it is important to maintains such condition in the lab.
3) Enzyme lipase catalyse the hydrolysis of fats results in the formation of fatty acids so there is increase in the acidity of the reaction.Increase in the acidity of the reaction is indicator of the hydrolysis of fat.
4)Protease enzyme catalyse the hydrolysis of proteins and thus acidity of the solution is increased due to digestion of proteins.Increase in the acidity of the reaction shows protease activity.
5) Benedict test is used to determine the presence of reducing sugars such as fructose, glucose, maltose and lactose.Formation of raddish precipitate indicate +ve Benedict test .Benedict test identify free ketone or aldehyde functional group.
6) Hydrolysis of urea is catalysed by urease enzyme and result in the formation of ammonia and carbamate. On further hydrolysis carbamate produces another ammonia molecule and carbonic acid. So production of ammonical smell indicate that urea has been hydrolysed.
7) Urease activity tends to increase the pH of its environment as it produces ammonia this is basic molecules. So ammonia is that molecule that is responsible for this observation.
8) Catalase enzyme catalyse the breakdown of hydrogen peroxide into water and oxygen as effervescence in nearly all living organism. This effervescence indicate this enzyme activity.
