Catalysis by the purified aldolase described in Question 2 was studied in at several temperatures (data shown below. Samples were taken from enzyme mixtures that had been incubated at the indicated temperatures for the indicated times and their catalytic activities were determined: 100 degrees C (, 105 degrees C (»), 110 degrees C (A), and 120 degrees C ( ). A relative activity of 100% was defined as the activity measured without any high-temperature incubation. Based on these data, what is unusual about the purified aldolase? In this study, the molar elipticity at 222 nm (an indicator of protein structure) was measured for a protein solution at various temperatures. Based on these data and those

Explanation / Answer

Please find the answer below:

An enzyme is extracted from a crude sample and purified by various methodologies for its enrichment. According to the image and the data, it can be clearly seen that there occurs enrichment of the target protein after various extraction processes. However, it must be carefully noted here that the yield of the enzyme after anion exchange chromatography was significantly high and pure which did not change much even after gel filtration chromatography. Thus, it can be clearly stated that anion exchange chromatography was the most efficient step in purification and enrichment of the protein.

The graph shows that when decrease in temperature takes place, the specific activity of the enzyme also decreases. This is contrary to the normal notion and scientific finding that being a protein by nature, an enzyme should degrade at high temperatures. However, this purified enzyme shows opposite results and works better at high temperatures of 100 degree centigrade and above.

According to the information, it can be clearly seen from the data and the images that the enzyme isolated was highly stable at high temperatures and purification and enrichment was performed at high temperatures only. Thus, the graph shown (last graph) definitely depicts increase in specific activity of the enzyme at high temperatures.
