Question

This Question comes from : Lab. Orgo I EXTRACTION of Coffeine from Tea Leaves Tea leaves contains: cellulose, caffeine, tannins & chlorophyll.
Using Thin Layer Chromatography: a) what is the stationary phase in this experiment b) what is the mobile phase in this experiment c) If a polar solvent is used, would the retention factor (Rf) of a compound be larger or smaller than a non-polar solvent? d) Would it be possible for retention factor (Rf) be greater than 1? Explain.
Equation: Rf=distance moved by sompound spot / distance moved by the solvent front This Question comes from : Lab. Orgo I EXTRACTION of Coffeine from Tea Leaves Tea leaves contains: cellulose, caffeine, tannins & chlorophyll.

Explanation / Answer

a) Stationary phase in thin layer chromatography is always silica gel (in some cases Alumina)

b) Mobile phase is a mixture of Hexane and ethyl acetate (Mixture of non-polar and polar systems)

c) Equation: Rf=distance moved by compound spot / distance moved by the solvent front

In polar system compound spot moves larger distance than non-polar system hence

Rf factor is of a compound is larger in polar systems than non-polar systems.

d) It’s not possible for Rf greater than 1. It’s impossible for the compound spot to cross the distance travelled by the solvent. The distance travelled by compound is always less than the distance travelled by the solvent. Hence due to this reason Rf factor is not greater than 1
