Question
The 1D 1H-NMR and corresponding 2D COSY spectra of the tetrapeptide Val-Gly-Asp-Glu at pH 3.5 in H20 at 25°C.
Draw the chemical structure of the peptide and indicate the expected 1 H spin systems. The "fingerprint" region of the COSY spectrum of a peptide or protein contains crosspeaks between HN and H protons
Why do we see crosspeaks from only three spin systems (or amino acids) in the fingerprint of Val-Gly-Asp-Glu? Connect the spin systems from the 4 residues of the peptide in the COSY spectrum (suggestion - use colored pens/pencils).
Interpret its 1D and 2D spectra by assigning as best possible all of the peaks to the appropriate protons in the peptide.
Explanation / Answer
Nuclear magnetic resonance spectroscopy is called as NMR-Spectroscopy . It is used for the determining the structure of organic compounds. Of all the spectroscopic methods,it is only one for which a complete analysis.Larger amount of sample required for mass spectroscopy when compared to NMR.
