Interpret these results in regards to the expression of the PAL5 gene of these two plants. In this case, a-tubulin is the housekeeping gene. In this case, the tomatoes A and B were treated with 20X times the normal NaCl found in the soil. Calculate the upregulation using the double delta Ct. – be sure to show all of your work. Explain what the results mean biologically.

For tomato A,
Delta Ct (for contro/basel) = Ct for PAL5 base - Ct for a-tubulin
= 36-33= 3
For tomato B ,
Delta Ct for control/ base = 34-32=2
For tomato A,
Delta Ct after experiment i.e. after adding 20X NaCl,= Ct for PAL6 experimental- Ct for a-tubulin experimental
= 32 - 35= -3
For tomato B,
Delta Ct for experimental = 31-32 =-1
Now, double delta Ct = delta Ct for experimental- delta Ct for control
Hence for tomato A,
Double delta Ct =-3-3 = -6
For tomato B,
Double delta Ct =
Double delta Ct = -1-2 = -3
Expression fold change = 2-double delta Ct value
Hence expression fold change of PAL5 in tomato A
= 2-(-6) = 26= 64 i.e. PAL5 is 64 times upregulated when the plant is treated with 20X NaCl.
In tomato B,
Expression fold change = 23= 8
Hence, PAL5 is produced 8 times more when the plant was treated with 20X NaCl as compared to when given normal NaCl treatment.
So, we can say that NaCl induces the expression of PAL5.
