a. dark purple

b. colorless

c. blue

d. red

2. What color do you expect mycobacterium smegmatis to be after performing the acid-fast stain?

a. blue

b. dark purple

c. colorless

d. red

3. What is the purpose of heating the slide during the staining process?

a. Eliminates contaminating bacteria

b. Allows for further preparation of the stain through the lipoidal wall and into the cytoplasm

c. Kills the bacteria and adheres it to the slide.

d. Separates the bacteria from one another for better visualization.

Explanation / Answer

Acid fast(AF) staining uses dye which has affinity for lipids which are found in the cell walls of my do bacteria and similar species. This stain is red in colour and hence the acid fast bacteria retain this stain and appear red. The counter stain used is methyl blue. The non acid fast bacteria lose the red colour and stain blue.

1) c blue ( E coli is AF negative)

2)d Red( AF positive)

3)c kills bacteria and adheres them to the slide.
