Borrelia burdorferi is an gram negative bacteria, and a spirocheate. They are thin, helical, highly motile and highly wound. They are visible through Giemsa staining. When serial dilutions are done and each one are plated, it is easy to count the number of bacteria because, there will be decrease in the number of bacterial cells, with increase in dilution. For higher dilution there will be minimum number of cells, that are easy to enumertae. Inb this way dilution technique is used to calculate the number of cells. Colony forming unit is the measure of number of viable cells in the plate. Bacillus anthracis can be the other microbe. Hematoxylin and methylene blue can be used as stains in the basic staining.
