Question

The structure shown below is a protein Richardson representation. Key components of the structure are labeled with the numbers 1 through to 5. Answer A, B, and C. What is the major type of secondary structure shown in the representation? Which label (1, 2, 3, 4, or 5) shows the N terminus of the protein? What parts of a protein structure would you expect to be most conserved during the course of evolution? You may refer to the structure shown here in your answer, however your answer should be applicable to any protein structure.

Explanation / Answer

Q 5. A . Here beta pleated sheet is shown in the representation.

Beta sheets consist of beta strands connected laterally by at least two or three backbone hydrogen bonds, forming a generally twisted, pleated sheet. A -strand is a stretch of polypeptide chain typically 3 to 10 amino acids long with backbone in an extended conformation. Antiparallel -sheets are slightly more stable than parallel ß sheets because the hydrogen bonding pattern is more optimal.

B. In this diagram label 2 shows the N terminus of the proein.

C. A protein domain is a conserved part of a protein sequence that means it conserved during the course of evolution. Protein domains are distinct functional and/or structural units in a protein. Usually they are responsible for a particular function or interaction, contributing to the overall role of a protein. Domains may exist in a variety of biological contexts, where similar domains can be found in proteins with different functions.

In this diagram, SH3 (Src homology 3) domain is shown. This domains are small domains of around 50 amino acid residues that are involved in protein-protein interactions. They occur in a diverse range of proteins with different functions, including adaptor proteins, phosphatidylinositol 3-kinases, phospholipases and myosins.
