Question
The data below were collected for an enzyme-catalyzed reaction. Determine V_max and K_m for this reaction. This graph is provided for your use only. It will not be graded. (It is not necessary to use a graph to answer.) Click on the graph to activate the Tools menu. Choose "Point" and then click a place on the graph to place the point. To move a point, choose "Select," and then drag the point. Use "Select" to scroll over a point to see its coordinates, or to see the equation of a line. Use the option "Line" to draw a line.Explanation / Answer
v = Vmax / (1 + (Km/[S]))
use any 2 rows of table and form 2 equations.
I am using row 1 and row 3
from row 1
2 = Vmax / (1 + (Km/0.25))
2 + 8 km = Vmax ....eqn 1
from row 3
5 = Vmax / (1 + (Km/1))
5 + Km = Vmax ....eqn 2
solve eqn 1 and eqn 2
2 + 8 km = 5 + Km
7 Km = 3
Km = 3/7 = 0.43 mM
Put value of Km in eqn2
Vmax = 5 + Km = 5 + 0.43 =5.43 mMs-1
Answer:
Vmax =5.43 mMs-1
Km = 0.43 mM
