Question
A series of Equilibrium dialysis experiments were set up where a fixed concentration and volume of the protein is contained within a sample chamber and dialyzed in the presence of ligand until an equilibrium is reached against a blank solution in the assay chamber. This allows the "free" ligand to reach an equilibrium between both sides while of course the "bound" ligand is contained entirely inside the sample chamber. Concentrations were quantitated by radiolabeling. Micromolar not milimolar
The following data was obtained:
Protein concentration in experiment: 0.918 mg/L
Polypeptide Molecule Weight of Protein: 55,800
Monlecular Weiht by Gel Filtration (no subunit dissociation) : 224, 000
Question: Calculate the Kd for ligand binding using a simple hyperbola fit
* I do not have logger pro and I can't figure out how to do this in xcel, please help!
Explanation / Answer
You will have to plot the data obtained(the set of all the points). Store the data column wise. Then select insert graph in excel(appears under the insert tab, then graph). Without the complete data, I won't be able to solve the problem, since I will have to plot in excel too.
