Question

Shown below is the absorbance spectrum of CBBG in the presence of a large excess of BSA. The x-axis shows wavelength (in nm) and the y-axis is absorbance – the y-axis is arbitrary since the absorbance depends on the concentration. You’ll notice that the maximum of this absorbance spectrum occurs at 595 nm, which is where you measured the absorbance of the complex. Why do molecules absorb light? Why are only certain wavelengths absorbed? Why was the molar extinction coefficient determined experimentally? What does this value depend on?

1.0 T 0.5 0.0 400 500 600 700 800

Explanation / Answer

In molecules there are many enrgy levels that can be occupied by electrons. Molecules absorbs light toget exceited to higher energy level
--------------------------------------------------------------------------------------
These excitated states are fixed and hence the difference in energy between these states are fixed. Molecules can absorb light of these corresponding energies only.Since E =h*c/wavelength, molecules can absoeb certain wavelength only.
-----------------------------------------------------------------------------------------
Molar extinction coefficient is an intrinsic property. It depends on the solution that you make. There is no preknown value for it. So its value must be determined experimentally.
-----------------------------------------------------------------------------------------------------
Its value depends on chemical species in solution/molecules. It depends on chemical composition and chemical structure of material
Note: It doesn't depend on concentration
